Phenibut (β-phenyl-γ-aminobutyric acid HCl) is a neuropsychotropic drug that was developed and introduced into clinical practice in Russia in the 1960s. It is known for its anxiolytic and nootropic effects. Similarly to pregabalin and gabapentin, phenibut acts as a strong VGCC (Voltage Gate Calcium Channel) blocker at the α2 subunit. In addition to that, phenibut exerts its therapeutic effects via its GABA-mimetic action, primarily at GABA-B and, to some extent, at GABA-A receptors. In that aspect, the psychopharmacological activity of phenibut is similar to that of baclofen, a p-Cl-derivative of phenibut. Furthermore, phenibut antagonizes β-phenethylamine (PEA), a presumed endogenous anxiogenic. It also stimulates dopamine receptors, which most likely accounts for its focus enhancing properties.

Phenibut has been demonstrated to act as a neuroprotective agent in vitro, however the exact mechanism of action in this case remains unknown.

In Russia, clinicians prescribe phenibut for tension, anxiety, and fear relief, to improve sleep improve sleep in psychosomatic or neurotic patients.

It is also used as a pre- or post-operative medication, as well as in the treatment of disorders characterized by asthenia and depression, in post-traumatic stress, stuttering and vestibular disorders.

In recent years, phenibut has gained its recognition worldwide as an anxiolytic and nootropic agent.

According to anecdotal evidence, low-dose phenibut is particularly useful for achieving a clear-headed state of mind, enhanced focus and motivation, mood improvement, combating stress and social anxiety. Phenibut seems to have its unique profile that encourages socialization in a manner distinct from benzodiazepines (which merely increase socialization through disinhibition), in that it directly increases the urge to communicate with others.

It is important to keep in mind that while phenibut is energizing and tends to increase libido at lower doses, it is highly sedative at higher doses.

Bodybuilders and athletes use phenibut before resistance training, as studies show that raising GABA levels leads to increased secretion of human growth hormone (HGH).

Dosage

With phenibut, it’s important to start low, at 200-300 mg per day. Phenibut is best absorbed on empty stomach. It takes a longer while to work (up to 3 hours in some users), while its effects can be long-lasting (up to 16 hours), which is why it is of great importance to refrain from premature redosing.

Since tolerance to its effects may develop, it is safe not to take phenibut more than three times a week and never exceed 2-3 grams per day.

Phenibut is not meant for long-term daily use, as in such cases dependence may develop. Withdrawal symptoms include anxiety, agitation, depression, cognitive deficits, fatigue and depersonalization. In more severe cases, vomiting, heart palpitations, tremor, insomnia, and visual and auditory hallucinations might be experienced.
