Selank is a nootropic, anxiolytic peptide-based pharmaceutical created by the Institute of Molecular Genetics of the Russian Academy of Sciences. Selank is a heptapeptide characterized by the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro (TKPRPGP). It is a synthetic counterpart of the immunomodulatory peptide tuftsin, hence emulating its numerous actions.
It has been demonstrated to regulate the expression of pro-inflammatory cytokines (TNF-α, IL-6), and influence the equilibrium of T helper cell cytokines. What is more, it has been shown to affect the levels of monoamine neurotransmitters and stimulate serotonin metabolism. Selank has been shown to swiftly increase the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus of rats. By increasing GABA-B receptors, it reduces anxiety without sedation. Furthermore, it is hypothesized to upregulate 5-HT1A receptors. Selank and its similar peptide medication, Semax, have been shown to inhibit enzymes that degrade enkephalins and other endogenous regulatory peptides, potentially contributing to their effects. It has also been shown to influence the activity of carboxypeptidase H and phenylmethylsulfonylfluoride-inhibited carboxypeptidase in rat nervous system tissue.
Moreover, Selank has been found to exhibit antidepressant-like effects in animal models of depression and anhedonia. In clinical trials, the medication has demonstrated a prolonged nootropic and anxiolytic effect, beneficial for the treatment of generalized anxiety disorder (GAD). With its efficacy is on par with that of diazepam, selank possesses an advantage over conventional anxiety therapies, including benzodiazepines and barbiturates, as it lacks sedative effects, is devoid of addiction or withdrawal issues, and does not impair cognition long-term.
Conversely, it enhances memory and reduces stress-induced cognitive decline.
Selank users typically relate experiencing calm focus, reduced social anxiety, and verbal fluency improvements with this compound. Its chronic use lowers baseline anxiety and improves stress resilience. Some neurodivergent individuals report being less susceptible to overstimulation and seem to behave in a more adequate manner in social situations.
